Integration of Single-Cell RNA Sequencing and Bulk RNA Sequencing Reveals That TAM2-Driven Genes Affect Immunotherapeutic Response and Prognosis in Pancreatic Cancer

Tumor-associated macrophages M2 (TAM2), which are highly prevalent infiltrating immune cells in the stroma of pancreatic cancer (PC), have been found to induce an immunosuppressive tumor microenvironment, thus enhancing initiation and progression. However, therapy response prognostic significance regulatory genes associated with TAM2 PC currently unknown. Based on TCGA transcriptomic data single-cell sequencing from GEO database, we identified TAM2-driven using WGCNA algorithm. Molecular subtypes based were clustered ConsensusClusterPlus The study constructed a model through Lasso-COX regression analysis. A total 178 samples obtained by accessing accurately categorized into two molecular subtypes, including high-TAM2 infiltration (HMI) cluster low-TAM2 (LMI) cluster. HMI exhibits poor prognosis, malignant phenotype, immune-suppressive cell infiltration, resistance immunotherapy, high number genetic mutations, while LMI is opposite. composed six hub degree accuracy predicting prognosis patients serves as independent risk factor. induction was employed means verifying these gene expressions, revealing significant up-regulation BCAT1, BST2, MERTK cells. In summary, immunophenotype offers foundation for clinical management PC. core genes, MERTK, involved regulating progression polarization, potential targets therapy.
